چکیده

The reaction of several plant chlorophyll-protein complexes with NaBH, has been studied by absorption spectroscopy. In all the complexes studied, chlorophyll b is more reactive than Chl a , due to preferential reaction of its formyl substituent at C-7. The complexes also show large variations in reactivity towards NaBH, and the order of reactivity is: LHCI > PSI1 complex > LHCII > PSI > P700 (investigated as a component of PSI). Differential pools of the same type of chlorophyll have been observed in several complexes. Parallel work was undertaken on the reactivity of micellar complexes of chlorophyll a and of chlorophyll b with NaBH, to study the effect of aggregation state on this reactivity. In these complexes, both chlorophyll a and b show large variations in reactivity in the order monomer > oligomer > polymer with chlorophyll b generally being more reactive than chlorophyll a. It is concluded that aggregation decreases the reactivity of chlorophylls towards NaBH, in vitro, and may similarly decrease reactivity in naturally-occurring chlorophyll-protein complexes.

The size and detergent binding of membrane proteins.

Sucrose density gradient centrifugation has been used to measure the binding of Triton X-100 above its critical micellar concentration to a variety of purified membrane and non-membrane proteins. In addition, binding studies were done on the three proteins below the critical micellar concentration of detergent to distinguish between the interaction of proteins with detergent monomers and deter...

The binding of detergents to lipophilic and hydrophilic proteins.

The lipophilic protein moieties of human low density lipoprotein, Semliii Forest virus envelope, and human erythrocyte stroma were obtained lipid-free with the use of sodium deoxycholate treatment and gel filtration in deoxycholatecontaining media. Detergent binding was determined by subjecting the proteins to gel filtration in the presence of micellar concentrations of radioactive deoxycholate...

A Surfactant-Induced Functional Modulation of a Global Virulence Regulator from Staphylococcus aureus.

Triton X-100 (TX-100), a useful non-ionic surfactant, reduced the methicillin resistance in Staphylococcus aureus significantly. Many S. aureus proteins were expressed in the presence of TX-100. SarA, one of the TX-100-induced proteins, acts as a global virulence regulator in S. aureus. To understand the effects of TX-100 on the structure, and function of SarA, a recombinant S. aureus SarA (rSa...

Simultaneous Extraction, Enrichment and Removal of Dyes from Aqueous Solutions Using a Magnetic Aqueous Micellar Two-Phase System

The magnetic aqueous micellar two-phase system (MAMTPS) has the advantages combined of magnetic solid phase extraction (MSPE) and aqueous micellar two-phase system (AMTPS). Thus, MAMTPS based on Fe3O4 magnetic nanoparticles (MNPs) and a nonionic surfactant Triton X-114 (TX-114) was developed for the extraction, enrichment and removal of three dyes (Congo red, methyl blue, and methyl violet) fro...

Membrane-Surfactant Interactions

Complex I11 (ubiquinol-cytochrome c reductase) was purified from beef heart mitochondria in the form of protein-phospholipid-Triton X100 mixed micelles (about 1:SO:lOO molar ratio). Detergent may be totally removed by sucrose density gradient centrifugation, and the resulting lipoprotein complexes retain full enzyme activity. In order to understand the role of surfactant in the mixed micelles, ...
